Design Notes

The original pyruvate kinase gene contained two PstI restriction sites beginning at bp 550 and 908. These restriction sites were removed by either 1) site directed mutagenesis or 2) the purchasing of designed oligos that were stitched together using Gibson Assembly.


Source

The part was derived from an existing previously constructed plamid in E. coli, carrying the rabbit muscle pyruvate kinase.
